QSP Approaches to Determine Best-in-Class Properties for Targeted Anabolic Growth Factor to Arthritic Joints

Abstract

We used Quantitative Systems Pharmacology (QSP) approaches to determine best-in-class properties for targeted anabolic growth factor to arthritic joints. The QSP model was used to determine what properties of the therapeutic would be predicted to have the greatest effect on the target activity at a maximal dose. Example properties include drug binding affinity, half-life, and target ligand binding affinity, target and ligand expression levels, target internalization kinetics, target expression, and cell numbers. The QSP model was also used to systematically map out drugs and target parameter space to identify tradeoffs between target and drug properties, and knowledge gap analysis and experiment prioritization by identifying sensitive biological parameters.
